Lancashire skipper Steven Croft was left ruing his decision to bowl first as Petersen and Gareth Rees (76) re-wrote the record books with a stand of 199 for the first wicket.
The pair were devastating throughout, with Petersen cracking nine fours and ten sixes during his fantastic knock.
The South African was eventually dismissed by Tom Smith having fired his side to 239 for two at the start of the 28th over.
Chris Cooke and Mark Wallace followed their skipper back to the pavilion within the space of two overs but a magical 19-ball 51 from Stewart Walters together with a quickfire 31 from Graham Wragg maintained the momentum as Glamorgan posted 328.
Needing nearly ten an over, Lancashire were under the cosh from the start. Matters didn't improve when Tom Smith was out in the first over, looking to sweep.
Steven Moore (57 off 38) and Croft (53 off 42) briefly threatened with a fourth-wicket stand of 94 but with wickets falling all too regularly, the Lightning charge soon petered out.
