Prior pulled up while going for a single on day four of the rain-reduced match, which ended in a draw, and did not take the field with the gloves when Australia A batted.
Joe Root, who had been batting with Prior when he became injured, took the gloves instead, as coach Andy Flower confirmed: "He's tweaked something in his calf. He will have to have a scan."
If Prior is ruled out of the first Test in Brisbane on 21 November, this will likely mean Jonny Bairstow will keep wicket as he does for Yorkshire.
