On-field officials Marais Erasmus and Ian Gould, television umpire Chris Gaffaney and match referee Jeff Crowe reported the possible transgression at the conclusion of the fourth ODI against England at Lord's last week.
Senanayake is required to undergo testing within three weeks – and is allowed to continue bowling until the results are revealed.
The 29-year-old is the visitors' leading wicket-taker in the ongoing five-match series, which is currently locked after two wins apiece. He has claimed eight victims in four innings, at an average of 12.50 and 3.82 economy rate.
The fifth and final ODI will get underway at Edgbaston in Birmingham on Tuesday. Two Tests will follow.
